MOGAT2 encodes monoacylglycerol O-acyltransferase 2 (MGAT2), a rate-limiting enzyme in intestinal triglyceride (TG) synthesis. It catalyzes the acylation of monoacylglycerol with acyl-CoA to form diacylglycerol, a core step in dietary fat absorption by enterocytes.
Dysregulation of MOGAT2 is associated with obesity, hyperlipidemia, and type 2 diabetes. Its inhibition reduces intestinal TG uptake, making it a potential target for metabolic disorders. Preclinical small-molecule inhibitors have shown efficacy in lowering plasma TG levels and body weight, though clinical development remains in early phases.
