NEFM

NEFM encodes the neurofilament medium polypeptide (NF-M), a core component of the neuronal cytoskeleton. As an intermediate neurofilament chain, it assembles with light (NF-L) and heavy (NF-H) chains to form heteropolymers, maintaining axonal structure, regulating axon diameter, and supporting efficient nerve impulse conduction. Common aliases include NF-M and neurofilament protein M.
Abnormal NF-M phosphorylation or aggregation is associated with neurodegenerative disorders like Alzheimer’s disease and amyotrophic lateral sclerosis (ALS), where misfolded NF-M disrupts axonal transport and neuronal integrity. Preclinical research explores small molecules and antibodies targeting NF-M misfolding, though no specific therapies are yet clinically approved.
