NNMT

NNMT encodes nicotinamide N-methyltransferase, a cytosolic enzyme that catalyzes nicotinamide methylation using S-adenosylmethionine as the methyl donor. It regulates NAD+ metabolism by diverting nicotinamide from salvage pathways, influencing cellular energy homeostasis and epigenetic regulation through methyl group transfer. This enzyme is linked to metabolic disorders, with overexpression in obesity and type 2 diabetes via NAD+ depletion and altered methylation patterns. Emerging preclinical studies target NNMT with small-molecule inhibitors to restore NAD+ levels, showing promise in improving insulin sensitivity and reducing hepatic steatosis in rodent models.
