PNPLA2

PNPLA2 encodes patatin-like phospholipase domain-containing protein 2, commonly known as adipose triglyceride lipase (ATGL). As the rate-limiting enzyme in triglyceride catabolism, it initiates the hydrolysis of stored triglycerides into diacylglycerol and free fatty acids, critical for energy mobilization from adipose tissue and lipid homeostasis in the heart, skeletal muscle, and liver.
ATGL activity is regulated by cofactors (e.g., CGI-58/ABHD5) and inhibitors (e.g., G0S2). Loss-of-function mutations cause neutral lipid storage disease with myopathy (NLSDM), while overactivity links to insulin resistance and metabolic syndrome. Preclinical small-molecule modulators—including inhibitors for metabolic disorders and activators for cachexia—are under investigation, though clinical translation is early.

Overlay Peak curve showing HepG2 cells stained with CSB-RA111385A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*10⁶cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*10⁶cells) used under the same conditions. Acquisition of >10,000 events was performed.
