Semaphorin-4C (SEMA4C), also known as Sema4C or CD100-like protein, is a member of the semaphorin family of signaling molecules. It mediates cell-cell communication by binding to receptors like Plexin B2, activating Rho GTPase pathways that regulate cytoskeletal rearrangement, cell migration, and axon guidance during development.
Aberrant SEMA4C expression is linked to malignancies including breast and colorectal cancer, where it promotes tumor invasion and metastasis. Preclinical studies are exploring targeted therapies such as anti-SEMA4C monoclonal antibodies to inhibit its pro-tumorigenic effects, though no such drugs have entered clinical use yet.
