SKP2, also known as S-phase kinase-associated protein 2, has alternative names including p45, FBXW2, and SKP2. SKP2 is a component of the E3 ubiquitin ligase SCF complex, comprising an F-box sequence, linker sequence, C-terminal region, and ten leucine-rich repeat (LRR) domains, where the LRRs play a crucial role in substrate recognition. SKP2 can induce ubiquitination at the K48 residue, leading to proteasome-mediated hydrolysis of labeled substrates, affecting the stability of various cell cycle-related proteins such as p27, FOXO1, and CARM1. By controlling the stability of cell cycle-related proteins, SKP2 participates in cell cycle regulation, proliferation, migration, and tumor development, with its expression levels closely related to the occurrence, progression, and prognosis of multiple tumors.

Immunofluorescence staining of HepG2 Cells with CSB-RA155372A0HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeated by 0.2% TritonX-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4℃. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Rabbit IgG (H+L).

The cells were fixed with 70% Ethylalcohol (18h) and then incubated in 10% normal goat serum to block non-specific protein-protein interactions followedby the antibody (1µg/1*106 cells) for 1 h at 4℃.The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 30min at 4℃.
