STUB1, full name STIP1 homology and U-box containing protein 1, is commonly known as CHIP with aliases like HSPABP2 and UBOX1. As an E3 ubiquitin ligase, it mediates K48-linked ubiquitination to drive proteasomal degradation of target proteins. It negatively regulates IFNγ signaling by degrading IFNGR1-JAK1 complex, and modulates mTORC1 pathway via p62 ubiquitination, also regulating antiviral RNAi through Dicer and AGO2 degradation.
STUB1 mutations cause spinocerebellar ataxias such as SCA48 and SCAR16, and its dysregulation links to cancers, rheumatoid arthritis and osteomyelitis. Preclinical studies show STUB1 inhibition enhances CD8+ T cell anti-tumor activity by upregulating IL-27Rα. STUB1-CHIC2 pathway inhibitors and STUB1-deficient engineered T cells are promising for cancer immunotherapy, with ongoing research into its clinical translation.

Immunofluorescence staining of MCF-7 cell with CSB-RA058285A0HU at 1:50 , counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
