SUCNR1

SUCNR1, or Succinate Receptor 1 (common alias GPR91), is a G-protein coupled receptor (GPCR) that functions as an extracellular succinate sensor. Succinate—a key tricarboxylic acid (TCA) cycle metabolite—accumulates outside cells during metabolic stress or hypoxia; SUCNR1 mediates downstream responses in tissues like the kidney, liver, and immune cells.
Activation of SUCNR1 triggers Gq/11 and Gi/o signaling pathways, leading to calcium mobilization, MAPK activation, and pro-inflammatory cytokine release. It is linked to renal ischemia-reperfusion injury, renal cell carcinoma, type 2 diabetes, and inflammatory disorders. Preclinical studies are exploring SUCNR1 antagonists as potential therapies, though no drugs have advanced to clinical trials yet.
