TIGIT (T cell immunoreceptor with Ig and ITIM domains) is an immune-regulatory receptor, consisting of an extracellular Ig-like V-type domain, a transmembrane region, and an intracellular domain containing an Immunoreceptor Tyrosine-based Inhibition Motif (ITIM). It is typically expressed on T cells, Natural Killer (NK) cells, and certain dendritic cells. Its primary role is to regulate the activity of T cells and NK cells within the immune system. TIGIT interacts with its ligands CD155 (also known as PVR) and CD112 (also known as PVRL2), thereby inhibiting the activation and cytotoxicity of immune cells. Through its intracellular ITIM domain, TIGIT recruits inhibitory signaling molecules such as tyrosine phosphatases, thus suppressing cell signaling pathways and reducing the immune cells' aggressiveness towards pathogens and tumor cells. This regulatory function helps maintain immune balance and reduce the occurrence of autoimmune diseases. Research on TIGIT also contributes to a deeper understanding of immune regulation mechanisms, potentially offering new insights into the treatment of other autoimmune diseases.
