TRIM63

TRIM63 encodes Tripartite Motif Containing 63, commonly known as MuRF1. As an E3 ubiquitin ligase, it mediates ubiquitination of sarcomeric proteins (e.g., myosin heavy chain) to target them for proteasomal degradation, playing a central role in skeletal and cardiac muscle atrophy.
Its activity is regulated by key pathways: FoxO transcription factors upregulate TRIM63 during atrophy, while mTOR signaling suppresses it. Dysregulation links to muscle wasting disorders (e.g., muscular dystrophy, cachexia) and cardiac dysfunction. Preclinical studies explore TRIM63 inhibitors as potential therapies to preserve muscle mass in these conditions.
