UCHL1

Ubiquitin C-terminal hydrolase L1 (UCHL1), also known as PGP9.5 or PARK5, is a core deubiquitinating enzyme (DUB) in the UCH family. It catalyzes ubiquitin cleavage from peptide/protein conjugates, maintaining intracellular ubiquitin homeostasis and regulating protein turnover via the ubiquitin-proteasome system (UPS). UCHL1 also modulates autophagic flux, linking it to cellular quality control pathways.
Familial Parkinson’s disease (PD) is associated with UCHL1 mutations, while its dysregulation contributes to Alzheimer’s disease and cancers like lung and colorectal carcinoma (overexpressed in tumors). Preclinical studies are exploring UCHL1 inhibitors for cancer and function-restoring modulators for neurodegenerative disorders, though no targeted therapeutics have entered clinical trials yet.
