Description

The LexA antibody is prepared through the immunization of rabbits with the peptide corresponding to amino acids 1-202 of the recombinant E.coli LexA repressor protein and further purification from rabbit antiserum by protein G. It is a polyclonal antibody and exists as an unconjugated IgG. Its purity reaches up to 95%. It only reacts with the LexA protein of E.coli-origin. And this anti-LexA antibody has been validated for use in ELISA and WB analyses. In E.coli, the transcriptional repressor LexA controls the SOS response, which is a coordinated cellular response to DNA damage. The LexA protein inhibits gene expression by binding as a dimer to a 16-mer consensus sequence CTG-N10-CAG named SOS boxes.

Target Background

Function
Represses a number of genes involved in the response to DNA damage (SOS response), including recA and lexA. Binds to the 16 bp palindromic sequence 5'-CTGTATATATATACAG-3'. In the presence of single-stranded DNA, RecA interacts with LexA causing an autocatalytic cleavage which disrupts the DNA-binding part of LexA, leading to derepression of the SOS regulon and eventually DNA repair. Implicated in hydroxy radical-mediated cell death induced by hydroxyurea treatment. The SOS response controls an apoptotic-like death (ALD) induced (in the absence of the mazE-mazF toxin-antitoxin module) in response to DNA damaging agents that is mediated by RecA and LexA.
Gene References into Functions
  1. The conformational flexibility of unbound LexA is the key element in establishing a co-ordinated SOS response. PMID: 21576225
  2. LexA regulated genes exhibit phenotypic heterogeneity as high level expression is observed in only a small subpopulation. PMID: 21070632
  3. NMR analysis of LexA catalytic domain in its active conformation PMID: 15929009
  4. oligomerization of LexA is proposed to be a possible regulation mechanism of the Escherichia coli SOS regulon in response to environmental conditions resulting in acidic intracellular pH. Structural changes on LexA repressor promoted by acidic pH. PMID: 16701697
  5. LexA binds to and represses the promoter of antC of coliphage N15. PMID: 17586637
