APEH

APEH encodes acylpeptide hydrolase (EC 3.4.19.1), commonly aliased as acylamino-acid-releasing enzyme (AARE) or ACY1. A serine protease, it catalyzes N-terminal acylpeptide removal from oligopeptides, supporting protein turnover and peptide processing in the cytoplasm and lysosomes. It aids oxidized protein degradation and antigen processing, linking to cellular stress responses.
Dysregulated APEH is associated with neurodegenerative diseases (e.g., Alzheimer’s, Parkinson’s) via impaired protein clearance and inflammatory disorders. It interacts with the ubiquitin-proteasome system and autophagy pathways. Early-stage drug research explores small-molecule inhibitors to modulate APEH activity, targeting toxic protein aggregates, though clinical trials remain preliminary.
