ASPN

Asporin, encoded by the ASPN gene (aliases: SLRR2B, ASPS), is a member of the small leucine-rich proteoglycan (SLRP) family. Its N-terminal aspartic acid-rich domain defines its name and mediates interactions with TGF-β and BMP ligands, inhibiting their signaling by blocking receptor binding. Expressed in cartilage, bone, and connective tissues, it regulates extracellular matrix (ECM) synthesis in chondrocytes, critical for joint homeostasis.
Aberrant ASPN activity links to osteoarthritis (OA): the D14 allele (14 aspartic acid repeats) increases knee OA susceptibility and severity. It modulates TGF-β/BMP pathways involved in bone remodeling and fibrosis. While no targeted drugs are approved, preclinical studies explore asporin neutralizing agents to restore ECM balance in OA, leveraging its role as a key regulator of degenerative joint disease progression.
