CUL5

CUL5, also known as cullin 5, is a scaffold protein that forms the core of the cullin-RING ubiquitin ligase (CRL) complex, specifically CRL5. It functions by recruiting substrates for ubiquitination and subsequent degradation by the proteasome. CUL5 plays a critical role in regulating cell cycle progression, apoptosis, and cell differentiation. It interacts with various substrates, including MCL1, which is involved in cell cycle regulation and apoptosis. CUL5 is implicated in the pathogenesis of B-CLL and other cancers, and its expression is often downregulated in various cancer cells. Research into CUL5 as a therapeutic target is ongoing, with inhibitors targeting the CUL5-RNF7-UBE2F ubiquitin ligase complex showing promise in combating cancer resistance.
