FTH1, Ferritin Heavy Chain 1, alias FHC, FTH, FTHL6, HFE5, PIG15, PLIF and so on. It is widely derived from animal and plant cells and is the main component of ferritin.
FTH1 can catalyze iron ions in the body into Fe3+ and store them to maintain normal iron homeostasis; it has ferrous oxidase activity, which promotes iron oxidation and cellular uptake of iron; it also possesses an anti-oxidative stress function, which is capable of degrading free radicals and effectively preventing oxidative damage caused by excess iron. FTH1 is involved in iron metabolism, including iron absorption, transport and utilization, and also participates in physiological processes such as immune response and cell proliferation, and has a protective effect on cells.
FTH1 is also associated with the development of various diseases, such as iron overload, neurodegenerative diseases, etc. Therefore, the regulation of FTH1 expression may be a potential therapeutic target for various diseases.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

Overlay Peak curve showing NIH/3T3 cells stained with CSB-RA159341MA1HU (red line) at 1:100. The cells were fixed in 4% formaldehyde (15min) and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-Rabbit IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
