GAD2

Glutamate decarboxylase 2 (GAD2), also known as GAD65 for its 65 kDa molecular weight, is a critical enzyme mediating the conversion of glutamate to γ-aminobutyric acid (GABA)—the central nervous system’s primary inhibitory neurotransmitter. It localizes to presynaptic terminals, supporting GABA synthesis for synaptic transmission and maintaining neuronal excitability balance.
Dysregulation of GAD2 links to type 1 diabetes (GAD65 autoantibodies are diagnostic markers), epilepsy, anxiety, and schizophrenia via impaired GABAergic signaling. Research advances include refining GAD65 autoantibody assays for early diabetes detection and exploring therapies to modulate GAD2 activity, aiming to restore GABA levels in neuropsychiatric disorders.
