IFNGR1

The protein encoded by IFNGR1 is interferon gamma receptor 1 (IFNGR1), alias CD119. A transmembrane glycoprotein, it heterodimerizes with IFNGR2 to bind IFN-γ, activating the JAK1/JAK2-STAT1 pathway. This mediates antiviral defense, immune cell activation, and regulation of inflammatory responses.
Defects in IFNGR1 cause Mendelian susceptibility to mycobacterial diseases (MSMD), increasing vulnerability to intracellular pathogens like Mycobacterium tuberculosis. Therapeutic R&D includes agonists to enhance immune function in infections and antagonists for IFN-γ-overactivated autoimmune disorders, with clinical applications still evolving.
