MAN1A2

MAN1A2 encodes Mannosidase alpha class 1A member 2, an endoplasmic reticulum (ER)-resident α-1,2-mannosidase with aliases like ERMAN1 and MAN1. It mediates the trimming of α-1,2-linked mannose residues from N-glycosylated nascent proteins, a critical step in the ER-associated degradation (ERAD) pathway—facilitating recognition and clearance of misfolded proteins to maintain ER homeostasis.
Dysfunction of MAN1A2 is linked to congenital glycosylation disorders (CDG) and malignancies. In cancer, aberrant MAN1A2 expression modulates cell adhesion and metastasis via altered glycoprotein processing. Preclinical studies explore MAN1A2 inhibitors as anti-cancer candidates, targeting tumor-specific glycobiology to impede invasive and metastatic phenotypes.
