POLR2A (DNA-directed RNA polymerase II subunit RPB1), also known as RPB1 or POLR2, encodes the largest catalytic subunit of RNA polymerase II (Pol II), responsible for synthesizing mRNA precursors in eukaryotes. This multi-domain protein contains a C-terminal domain (CTD) with heptapeptide repeats whose phosphorylation regulates transcription initiation, elongation, and termination by recruiting RNA processing factors. POLR2A is central to transcriptional regulation, DNA damage response, and cell cycle control through interactions with P-TEFb, Mediator complex, and DNA repair machinery. Mutations in POLR2A are implicated in neurodevelopmental disorders (e.g., HLD22) and cancers, particularly glioblastoma and melanoma, where truncating variants disrupt transcriptional fidelity. Therapeutically, POLR2A's vulnerability in hemizygous cancers is exploited by α-amanitin-based antibody-drug conjugates (e.g., AMT-562), while CTD phosphorylation modulators are under investigation for transcription-targeted therapies. Recent clinical trials (NCT04849994) explore POLR2A-directed agents in TP53-wildtype malignancies.

IHC image of CSB-RA018327A02phHU diluted at 1:100 and staining in paraffin-embedded human ovarian cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
Immunofluorescence staining of Hela cells with CSB-RA018327A02phHU at 1:100,counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4℃. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG (H+L).
