TIGAR, the TP53-induced regulator of glycolysis and apoptosis, alias FR2BP, C12orf5. it is a protein that plays an important role in cellular response to p53-related stress, encoded by the gene of human cells themselves. the mechanism of action of TIGAR is mainly by inhibiting the glycolytic pathway and increasing the flow of the pentose phosphate pathway, so that it can produce more NADPH to promote DNA repair and reduce the production of intracellular reactive oxygen species (ROS) to protect cells from oxidative stress-induced damage.TIGAR has a wide range of biological significance, and is involved in the regulation of cellular metabolism, redox homeostasis, cell cycle, apoptosis, and autophagy, etc.
The expression of TIGAR is closely related to the development of a variety of diseases, especially in the development of tumors. TIGAR contributes to tumor cell evasion from immune surveillance and enhances the invasive ability of tumor cells through antioxidant effects and regulation of autophagy. In addition, the abnormal function of TIGAR has been associated with certain cardiovascular and neurodegenerative diseases.
