TM4SF1, also known as Transmembrane 4 L six family member 1, is a member of the TM4SFs subfamily within the Tetraspanin superfamily, initially identified in 1986. Characterized by the typical Tetraspanin four-transmembrane structure, it features two extracellular loop regions, EC1 and EC2, with EC2 capable of binding to integrin α5β1 through cysteine residues, thereby influencing cell adhesion and motility. The C-terminal cytoplasmic region contains a PDZ binding domain that interacts with Syntenin-1 and Syntenin-2 proteins, mediating tumor cell proliferation and metastasis. TM4SF1 is overexpressed on the surface of various tumor cells, including lung, breast, liver, colorectal, and ovarian cancers. It forms complexes with lipid kinases and tyrosine protein kinases, such as DDR1, activating the PKC and JAK2-STAT3 signaling pathways and promoting cellular pseudopodia extension. Preliminary research suggests that TM4SF1 serves as a co-receptor for DDR1, a single-transmembrane receptor with tyrosine protein kinase activity, regulating downstream PKC/STAT3 signaling, thus influencing breast cancer metastasis to lung tissue. Additionally, TM4SF1 is an ADC target with a novel internalization pathway, indicating its potential application in the design and development of antibody-drug conjugates (ADCs). Drug development targeting TM4SF1 is still in its early stages, with no mature therapeutics available for clinical use. Therefore, the research on TM4SF1 remains profoundly significant and holds vast prospects.
