TNFRSF13B, also known as the tumor necrosis factor receptor superfamily member 13B, is commonly referred to as TACI. It is a type I transmembrane protein. TACI binds to both BAFF and APRIL, which are cytokines, and activates downstream signaling pathways such as the NF - κB pathway. This activation is crucial for B - cell survival, proliferation, and antibody production.
Dysregulation of TNFRSF13B has been associated with various diseases, including autoimmune diseases like systemic lupus erythematosus and immunodeficiency disorders. In drug development, targeting TACI is a promising strategy. Some monoclonal antibodies are under investigation to modulate its activity, aiming to correct the abnormal immune responses related to TACI - mediated pathways.
