Tumor necrosis factor receptor superfamily member 1A (TNFRSF1A), also known as CD120a or TNFR1, is a type I transmembrane glycoprotein with a molecular weight of 55 kDa. TNFR1 is a member of the tumor necrosis factor (TNF) receptor superfamily and contains three parts, including extracellular domain, transmembrane domain and intracellular domain. The extracellular domain contains 182 amino acids and consists of four cysteine-rich domains (CRD1-CRD2-CRD3-CRD4). Each domain contains 6 conserved cysteine residues, contains 3 N glycosylation sites, and its function is mainly to bind to the trimeric form of TNF; the transmembrane region is between the extracellular domain and intracellular domains, it is about 22 amino acids long; the intracellular domain is about 223 amino acids long. The N-terminal residue is leucine. The C-terminus contains a death domain (DD) composed of 80 conserved amino acid sequences, which is necessary for the transduction of cell death signals, and many functions of TNFR1 are related to this domain.
TNFR1 has binding activity to TNF-α and TNF-β, and there are many studies on the binding of TNFR1 to TNF-α. When TNF-α binds to TNFR1, on the one hand, it can mediate apoptosis signal, cause apoptosis, play an important role in anti-tumor and anti-viral infection, and also participate in the damage to own tissue cells in autoimmune diseases and sepsis . TNFR1 can also mediate cell activation and proliferation signals, induce the activity of nitric oxide synthase and IL8, activate nuclear transcription factor (NF-κB) and activator protein 1. TNFR1 also induces the expression of intercellular adhesion factor 1 and IL6. As a popular biological target, TNFR1 has a dual role. A large number of studies have confirmed that the expression of TNFR1 can promote or inhibit the apoptosis of tumor cells and participate in autoimmune diseases.

Tumor necrosis factor receptor superfamily member 1A is a protein in humans that is encoded by TNFRSF1A gene. TNFRSF1A is a receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Contributes to the induction of non-cytocidal TNF effects including anti-viral state and activation of the acid sphingomyelinase.
