C-C chemokine receptor type 9 (CCR9), also known as CD199, is a G protein-coupled receptor (GPCR) that binds its ligand CCL25 (thymus-expressed chemokine, TECK). Its activation via Gαi/o signaling triggers calcium mobilization, MAPK pathway activation, and chemotaxis of T cells to intestinal mucosa, regulating mucosal immune homeostasis.
CCR9 is implicated in inflammatory bowel diseases (IBD) like Crohn’s disease and ulcerative colitis, as its signaling drives pathological immune cell infiltration. It also links to enteropathy-associated T-cell lymphoma. Small-molecule antagonists such as vercirnon (CCX282-B) have reached phase III trials for Crohn’s disease, targeting to block CCR9-CCL25 interaction and reduce gut inflammation.
