CRTAM (CD355), a newcomer to the immunoglobulin superfamily (Ig-SF) and a member of the nectin-like protein superfamily, plays a pivotal role in the formation, response, and cellular interactions of the immune system as a cell-surface transmembrane protein. CRTAM is expressed on activated human NK cells, CD8+ T cells, and subsets of CD4+ T cells, participating in cell adhesion, polarity, and proliferation, while regulating lymphocyte functions such as promoting the adhesion and retention of TCD8+ cells within lymph nodes. CRTAM has been associated with various diseases, including inflammation, asthma, malignant mesothelioma, and certain neoplastic conditions. It can influence the polarization of CD4+ T cells, promoting their further differentiation and enhancing cytokine secretion capabilities. Additionally, CRTAM plays a crucial role in NK cell-mediated tumor cell killing during the immune response, presenting a potential tumor-targeting strategy. CRTAM interacts with Necl-2, promoting NK cell cytotoxicity and CD8+ T cell IFN-γ secretion in vitro, as well as NK cell-mediated rejection of Necl-2-expressing tumors in vivo. The expression of CRTAM is tightly regulated and primarily found on activated immune cells. In healthy individuals, CRTAM expression is low in peripheral blood mononuclear cells (PBMCs), but it is upregulated in PBMCs from asthmatic patients. Similar to IL-2, the CRTAM promoter is regulated by the AP-1 transcription factor. Upon activation, CRTAM stimulates CD8+ T cells to secrete IFN-γ and enhances the killing abilities of both CD8+ T cells and NK cells.
