DNASE1L3

Deoxyribonuclease 1 Like 3 (DNASE1L3), also known as DNase gamma, is a secreted endonuclease that cleaves chromatin DNA into mononucleosomes. It plays a critical role in clearing apoptotic cells and immune complexes, thereby maintaining immune tolerance by preventing self-DNA fragments from triggering autoimmunity.
Mutations in DNASE1L3 are strongly linked to systemic lupus erythematosus (SLE) and other autoimmune disorders, as defective enzyme function leads to accumulated self-DNA and autoantibody production. Current research focuses on recombinant DNASE1L3 or modulators to restore its activity, though clinical applications remain in preclinical stages.
