DRD5 is a G protein-coupled receptor (GPCR) highly expressed in the limbic system's neuronal populations, notably in the prefrontal cortex and hippocampus. This receptor modulates cognitive function, emotion, and motor control by binding to dopamine and activating G proteins, which in turn influence cAMP levels and ion channel activity. DRD5 is part of the dopaminergic system that regulates reward, motivation, and attention, and its dysregulation is implicated in neuropsychiatric conditions such as schizophrenia, Parkinson's disease, and addiction. As a key target for pharmaceuticals, DRD5 agonists may offer treatments for cognitive disorders and neurodegenerative diseases. Additionally, DRD5 has been shown to curb inflammation through the DRD5-ARRB2-PP2A axis by inhibiting the TRAF6-mediated NF-κB pathway, highlighting its role in neuroimmune modulation.
