EIF2AK2

EIF2AK2 encodes eukaryotic translation initiation factor 2-alpha kinase 2, commonly known as PKR. Activated by double-stranded RNA (dsRNA) from viral infections, it phosphorylates the translation initiation factor eIF2α, halting protein synthesis to restrict viral replication. It also mediates innate immune signaling, including interferon production, to combat pathogens.
Dysregulation of EIF2AK2 is linked to viral diseases (HIV, HCV), autoimmune disorders (systemic lupus erythematosus), and cancer (tumor progression via altered translation control). Preclinical studies explore PKR inhibitors to treat viral infections or modulate immune responses; some agents are being evaluated for anti-cancer potential by targeting its kinase activity.
