ENOX2

ENOX2 (Ecto-NOX disulfide-thiol exchanger 2), commonly known as tNOX (tumor-associated NADH oxidase), is a membrane-bound protein with dual oxidase and disulfide isomerase activities. It catalyzes NADH oxidation and protein disulfide bond rearrangement, supporting cancer cell proliferation and migration by maintaining redox homeostasis and driving cell cycle progression.
Overexpressed in breast, prostate, and colorectal cancers, ENOX2 correlates with tumor aggressiveness. Small-molecule inhibitors targeting its catalytic domain—such as synthetic flavonoids or vanadium complexes—are under preclinical investigation as potential anticancer agents, aiming to disrupt its role in sustaining aberrant cell growth and metastasis.
