FLG

Filaggrin (encoded by FLG), also known as FLG1, is a critical structural protein in the epidermis. Synthesized as profilaggrin in keratinocytes, it is cleaved into monomers during terminal differentiation, which aggregate keratin filaments into dense bundles—key for stratum corneum formation and skin barrier integrity.
FLG expression is regulated by epidermal differentiation pathways like Notch and Wnt. FLG mutations strongly associate with atopic dermatitis, ichthyosis vulgaris, and allergic diseases due to impaired barrier function. Current treatments include topical emollients for barrier repair; novel strategies target upregulating FLG or correcting mutations via gene editing.
