FLT3

FLT3, also known as CD135, a type III receptor tyrosine kinase, is key to hematopoietic stem cell and precursor B cell development. The FLT3 gene mutation is a common genetic alteration in acute myeloid leukemia (AML) patients and is linked to a poorer prognosis. These mutations lead to continuous autophosphorylation of the FLT3 receptor independent of FLT3 ligands (FL), resulting in the aberrant activation of downstream signaling pathways such as PI3K/AKT, RAS/MAPK/ERK, and STAT5. AML patients with these mutations face a grim prognosis and high relapse risk. FLT3 inhibitors offer a therapeutic approach by blocking FLT3's ATP-binding sites, halting cell cycle progression and inducing differentiation. Thus, FLT3 is pivotal in leukemia pathogenesis and a target for targeted therapies.
