GP5

Glycoprotein 5 (GP5), alias CD42d, is a platelet membrane glycoprotein that forms a functional complex with GP1b-IX. It mediates platelet adhesion to von Willebrand factor (vWF) on damaged vascular walls, triggering intracellular signaling cascades like Src family kinase activation to drive platelet activation and aggregation.
Dysregulation of GP5 contributes to bleeding disorders such as Bernard-Soulier syndrome, where impaired complex function reduces platelet adhesion. Drug development targets the GP1b-IX-V complex; candidates include monoclonal antibodies and small molecules to inhibit thrombotic events while mitigating bleeding risks, though clinical applications remain in early stages.
