HLA-E

HLA-E encodes the HLA class I histocompatibility antigen, alpha chain E, a non-classical MHC class I molecule. It presents leader peptides from classical HLA class I heavy chains to CD94/NKG2 receptors on natural killer cells and cytotoxic T lymphocytes, regulating immune surveillance.
HLA-E is critical for maintaining immune tolerance and antiviral immunity via the CD94/NKG2A inhibitory pathway. Dysregulation associates with cancer immune evasion and infectious diseases like HIV. Its role in immune checkpoint modulation makes it a potential target for immunotherapeutic development, though clinical agents are currently in preclinical stages.
