Recoverin, encoded by the RCVRN gene, is a calcium-binding protein predominantly expressed in retinal photoreceptors. Its full name is recoverin, with no widely recognized aliases. It regulates rhodopsin kinase activity in a Ca²⁺-dependent manner: low intracellular calcium levels (post-light exposure) cause recoverin to dissociate from rhodopsin kinase, allowing the kinase to phosphorylate rhodopsin and terminate light signaling—critical for GPCR-mediated phototransduction and visual adaptation.
Dysregulation of RCVRN is linked to retinal degenerative disorders like retinitis pigmentosa, where impaired recoverin function disrupts phototransduction and leads to photoreceptor cell death. Current research includes preclinical small-molecule modulators targeting recoverin’s calcium-sensing domain and gene therapy to correct RCVRN mutations, aiming to preserve retinal function.
