S1PR1

S1PR1, also known as sphingosine-1-phosphate receptor 1, is a G protein-coupled receptor expressed predominantly in endothelial cells. It binds sphingosine-1-phosphate (S1P) with high affinity and plays a crucial role in lymphocyte trafficking, vascular development, and endothelial integrity. S1PR1 activation is involved in various signaling pathways, including Gi/o, Gq, and G12/13, regulating cell proliferation, migration, and survival. The S1PR1 pathway is associated with numerous diseases, such as multiple sclerosis, systemic lupus erythematosus, and inflammatory bowel disease. Drug development targeting S1PR1 has led to the approval of fingolimod for treating multiple sclerosis, with ongoing research focusing on selective S1PR1 modulators for various autoimmune and inflammatory disorders.
