UAP1

UAP1 encodes UDP-N-acetylglucosamine pyrophosphorylase 1, an enzyme catalyzing UDP-N-acetylglucosamine (UDP-GlcNAc) synthesis—the key precursor for O- and N-linked glycosylation. Common aliases include UDP-GlcNAc pyrophosphorylase 1. It modulates glycosylation pathways critical for protein folding, cell adhesion, and signal transduction in proliferation and differentiation.
Dysregulated UAP1 is associated with cancers (e.g., colorectal, liver) via aberrant glycosylation driving tumor growth and metastasis. Drug development targets UAP1 to disrupt tumor-specific glycosylation; most candidates remain in preclinical stages, with studies exploring their potential to inhibit cancer progression by blocking UDP-GlcNAc production.
