CTPS1

CTPS1 encodes carbamoyl-phosphate synthetase 1, a mitochondrial enzyme central to the urea cycle. It catalyzes carbamoyl phosphate synthesis from ammonia, bicarbonate, and ATP, critical for ammonia detoxification in liver. Deficiencies cause hyperammonemia, while its upregulation in cancers supports pyrimidine synthesis via the de novo pathway.
CTPS1 interacts with the mTOR-HIF1α pathway, promoting tumor cell proliferation under nutrient stress. Inhibitors like N-(phosphonacetyl)-L-aspartate (PALA) target its enzymatic activity, showing preclinical efficacy in acute lymphoblastic leukemia and hepatocellular carcinoma models by disrupting nucleotide metabolism.
