RSPO1 gene is a member of R-spondins gene family, located at 1p34, full-length 23Kb, contains 9 exons and 8 introns, and can encode 263 amino acids. The R-spindins family is an important Wnt activator discovered in recent years. It consists of four secreted proteins (RSPO1-4) with 40%-60% sequence similarity and similar structural patterns. R-spondin proteins (RSPO1-4) are secreted proteins that efficiently sensitize cells to Wnt/β-catenin signaling and Wnt/PCP signaling.
The role of RSPO1 in sex development is to participate in and stabilize the Wnt/β-catenin signal transduction pathway. Normally, β-catenin in the cytoplasm is maintained at a low level by ubiquitin proteasome-mediated phosphorylation and degradation. In the classic Wnt/β-catenin signal transduction pathway, RSPO1, as one of the Wnt ligands, interacts with Frizzled ( FZD) receptors bind to the low-density receptor-related protein (LRP6) coreceptor, triggering intracellular signal transduction and initiating the accumulation of intracellular β-catenin. Activation of the Wnt/β-catenin signaling pathway is determined by cytoplasmic β-catenin levels. When the classical pathway is activated, through a series of complex processes, non-phosphorylated β-catenin accumulates in the cytoplasm to avoid being degraded, and then translocates into the nucleus, where it interacts with transcription factors such as T cell factor (TCF)/lymphoid enhancer factor (LEF) The complex structure is formed, the transcription of target genes is activated, and ovary formation is induced. The reduction of RSPO1 activity inhibits the Wnt/β-catenin signaling pathway, thereby enhancing the expression of SOX9. In the presence of SRY, the action of RSPO1 is inhibited, allowing testicular development.
