R-Spondin 1 (RSPO1) is a secreted glycoprotein with aliases including CRISTIN3 and RSPO. It potentiates Wnt/β-catenin signaling by bridging LGR4/5/6 receptors and RNF43/ZNRF3 E3 ubiquitin ligases, blocking ligase-mediated degradation of Wnt receptors (FZD/LRP5/6) to stabilize cytoplasmic β-catenin, which translocates to the nucleus and regulates target gene expression.
RSPO1 mutations are linked to 46,XX testicular disorder of sex development and primary ovarian insufficiency. Its overexpression drives Wnt pathway activation in colorectal and breast cancers. Preclinical inhibitors targeting RSPO1-LGR interaction are evaluated for Wnt-dependent tumors, while recombinant RSPO1 shows promise in repairing intestinal epithelial damage.
