CXCL12, also known as Thymic lymphoma cell-stimulating factor (Tlsf, TLSF-a, TLSF-b, Tlsfa, Tlsfb, TPAR1), is a small molecule cytokine that plays important roles in various physiological and pathological processes. It is secreted by multiple cell types and is particularly involved in embryonic development, hematopoiesis, angiogenesis, and inflammation. The mechanism of action for CXCL12 primarily involves binding to its high-affinity receptor, CXCR4, and activating downstream signaling pathways, such as the Gαi protein-activated PI3K/AKT and ERK/MAPK pathways, thus influencing cell proliferation, migration, and survival.
In addition to its chemotactic effects, CXCL12 is closely related to the occurrence, development, and prognosis of tumors. In cancer, the CXCL12/CXCR4 signaling pathway promotes tumor cell proliferation, motility, adhesion, invasion, metastasis, and secretion, affecting tumor progression. Furthermore, the CXCL12/CXCR4 signaling pathway plays a role in ischemic stroke by mobilizing endothelial progenitor cells to participate in repairing vascular endothelium and neural damage, promoting brain function recovery and improving quality of life. Therefore, CXCL12 is an important molecular target in cellular biology and disease research.
