Mucin16 (MUC16) is a glycoprotein antigen recognized by the mouse monoclonal antibody OC125, which was reported by American scientist Bast et al. in 1981 using the ovarian epithelial cancer cell line OVCA433 as the antigen. MUC16 is a transmembrane protein and a member of the Mucin (MUC) family. MUC16 is highly expressed in more than 80% of ovarian cancers and is also overexpressed in endometrial, pancreatic, colon, breast and lung cancer cells. The extracellular part of MUC16 can be cleaved and released into extracellular matrix or serum, namely CA125, a tumor marker commonly used in clinical diagnosis and prognosis detection of ovarian cancer.
MUC16 is a multifunctional macromolecule whose extracellular domain extends from the cell surface in ovarian cancer cells, providing an early point of contact for the soon-to-metastasize ovarian cancer cells and distant peritoneal mesothelial cells . Studies have shown that the extracellular domain of MUC16 can specifically bind to mesothelin, a glycoprotein expressed on the surface of normal peritoneal mesothelial cells, and also expressed in ovarian cancer cells and mesothelioma cells. , can promote cell adhesion to the peritoneum. The interaction of mesothelin with MUC16 mediates the adhesion between homotypic and heterotypic cells, allowing ovarian cancer cells to metastasize to the peritoneum through adhesion to mesothelial cells. The interaction of MUC16 with mesothelin is a mechanism that enables ovarian cancer cells to adhere to the peritoneal surface, and its extracellular hyperglycosylated random repeats enable the binding of lectin to MUC16, which may be the cause of ovarian cancer Another mechanism of cell adhesion to host tissue. It can be seen that through the combination with mesothelin and lectin, the extracellular domain of MUC16 may be involved in regulating cell adhesion and invasion, and plays an important role in the process of tumor cell metastasis.
