S100A4, also known as S100 calcium-binding protein A4, is a member of the S100 protein family encoded by the S100A4 gene. Commonly referred to as metastasin-1 (MTS1) or fibroblast-specific protein-1 (FSP1), this EF-hand calcium-binding protein regulates cellular processes including cytoskeletal dynamics, cell motility, and extracellular matrix remodeling. It exerts pleiotropic effects through interactions with intracellular targets like non-muscle myosin heavy chain and extracellular receptors such as RAGE (receptor for advanced glycation end-products), activating pathways including MAPK, NF-κB, and Wnt/β-catenin signaling. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
