SEMA4D Research Reagents

Axon guidance protein 4D (SEMA4D), also known as CD100, is one of the important members of the axon guidance protein N subfamily (SEMAs IV). SEMA4D is widely distributed and highly expressed in embryonic and adult tissues, including non-lymphoid and lymphoid tissues. In lymphoid organs, SEMA4D is mainly expressed on the surface of resting T cells, while its expression is lower on resting B cells and antigen-presenting cells (APCs).

SEMA4D was originally discovered as an axon guidance molecule affecting neural development. Recently, more and more studies have shown that SEMA4D is involved in a variety of important physiological functions of the immune and non-immune systems. Exogenous SEMA4D or soluble recombinant SEMA4D protein can promote the activation of B cells. Moreover, SEMA4D can promote tumor angiogenesis by mediating endothelial cell migration and lumen formation. Studies have shown that the promotion of tumor angiogenesis of SEMA4D is similar to that of other important angiogenesis molecules, such as vascular endothelial growth factor (VEGF), HCF, bFGF and so on. In addition, SEMA4D can also promote the invasion and metastasis of the above malignant tumor cells through the Met signaling pathway after binding to Plexin B1. Plexin B1 is one of the three recognized SEMA4D receptors, the other two are Plexin B2 and CD72. Among them, Plexin B1 has the highest affinity with SEMA4D, and is highly expressed in various solid tumors, such as breast cancer, colon cancer, liver cancer, pancreatic cancer and gastric cancer.
