CLDN4

Claudin-4 (CLDN4), a key member of the claudin family of tight junction proteins, is also referred to as CLD4. It mediates intercellular adhesion to maintain epithelial barrier integrity, regulating paracellular transport of ions and small molecules across epithelial layers.
Aberrant CLDN4 expression is associated with Wnt/β-catenin pathway activation, driving tumor progression in pancreatic, gastric, and ovarian cancers. Current drug developments include Claudin-4-targeted antibody-drug conjugates (ADCs) and combination therapies with immune checkpoint inhibitors, which are in preclinical or early clinical stages for treating CLDN4-overexpressing malignancies.
