What is GH1?

Growth hormone is a single peptide protein hormone in the growth hormone / prolactin family. It is secreted by the growth hormone secreting cells of eosinophilic granules in the anterior pituitary, so it is also called pituitary growth hormone.

GH1 is synthesized and released into the blood circulation by the pituitary gland. After binding to its receptor, it can stimulate the production of insulin-like growth factor Ⅰ (IGF-Ⅰ), thus promote cell proliferation, differentiation and inhibit cell apoptosis. Growth hormone plays an important physiological role in the growth and development of animals. In addition, GH1/IGF-I pathway also plays an important role in the occurrence and development of cancer.

The Structure of GH1

The human genome contains five GH-related genes: human growth hormone (hGH), human chorionic prolactin-A (hCS-A), human chorionic prolactin-B (hCS-B), Human chorionic growth prolactin analogues (hCS-L) and human growth hormone variants (hGH-V). These genes were mapped on chromosome 17q22-24, with a total length of 66.5 kb.

GH1 gene is located on human chromosome 17q24 and encodes GH, a single-stranded, non-glycosylated hydrophilic globulin containing 191 amino acids. 22 kDa hGH is the most abundant and main growth hormone in human body. The protein consists of four antiparallel α-helix structures that are necessary for GH to interact with its receptors to function.

Expression and regulation of GH1

GH1 gene is mainly transcribed and expressed in growth hormone-secreting cells of anterior pituitary, and a small amount is expressed in lymphocytes.

The secretion of growth hormone (GH) in the pituitary is controlled by two hypothalamic peptides: GH-releasing hormone (GHRH) and somatotropin release inhibiting factor (SRIF).

GHRH promotes the transcription and release of GH gene, and SRIF inhibits the secretion of growth hormone.

Ghrelin, which stimulates the secretion of growth hormone.

In addition, age, sex, diet, exercise, stress and other hormones also affect the secretion of growth hormone.

The Function of GH1

Human growth hormone has a wide range of physiological functions.

GH1 can promote the growth and development of the body by binding to the receptor. Mutations or deletions in the gene can lead to growth hormone deficiency and short stature.

Growth hormone also has a wide range of physiological effects on nervous system diseases, especially in nerve repair and nerve protection.

The role of growth hormone also includes stimulating the proliferation and differentiation of chondrocytes and increasing bone mineral density; increasing muscle mass through sarcomere proliferation and stimulating the body's immune system.

The excess and lack of GH1 can also cause some diseases.

Excessive GH1 may cause acromegaly. Growth hormone deficiency can cause growth hormone deficiency.

Applications of GH1 in Treatment

rhGH is used in the treatment of growth hormone deficiency, Turner syndrome (gonadal insufficiency syndrome) and renal insufficiency. Because of the regulatory effect of rhGH on immune system, it also has a broad prospect in organ transplantation, rheumatism, anti-infection, anti-aging and so on.

Currently available recombinant growth hormones include Nutropin (Genentech), Humatrope (Lilly), Genotropin (Pfizer), Norditropin (Novo), Saizen (Merck Serono) and Omnitrope (Sandoz).
