GHR is a gene encoding a protein named growth hormone receptor (also abbreviated as GHR). GHR protein belongs to the class I cytokine receptor family that is widely expressed in GH target cells. The first step for GH to act at the tissue and cell level is to bind to GHR on the surface of the target cell, and GHR mediates the signal transmission into cells to activate diverse signal cascades, leading to growth promotion, cell division, and regeneration.
GHR is a single-chain transmembrane glycoprotein consisting of three regions, including extracellular region (245aa), transmembrane region (25aa) and intracellular region (350aa). In the extracellular region, there are five conserved glycosylation sites, which are the sites for ligand binding. Near the cell membrane, there is a WSXWS-like motif in the extracellular domain , which is a conserved sequence consisting of 5 amino acid residues such as Trp-Ser-XXX-Trp-Ser (XXX represents any amino acid). This structure may play a role in stabilizing the spatial conformation during the binding of GH to GHR. In the intracellular region, two conserved sequences have been found, a sequence close to the cell membrane-sequence box I (Box1) consists of 8 amino acids and is rich in proline. Another segment-sequence box 2 (Box2) contains 15 amino acids and is 30 amino acid residues away from Box I.
GHR belongs to the cytokinin/hematopoietin receptor superfamily. When GH binds to GHR, GHR undergoes a dimerization process, and finally one GH binds to two GHRs. The signal transduction pathways mediated by GHR mainly include: tyrosine kinase (JKA2) system and protein kinase C (PKC) pathway. Additionally, emerging evidence also revealed that GHR can also signal through the IRS pathway. And nutritional status and endocrine factors such as GH also have regulatory effects on the expression of GHR.
