TNFR1 (Tumor Necrosis Factor Receptor 1), also known as TNF-R55 or CD120a, is a receptor protein that interacts with Tumor Necrosis Factor-Alpha (TNF-α) and plays a crucial role in various physiological and pathological processes. It consists of an extracellular domain, a transmembrane domain, and an intracellular domain. The extracellular domain binds to TNF-α, triggering signal transduction. The intracellular domain contains a Death Domain, which plays a critical role in initiating apoptosis (cell self-destruction) and other cell signaling pathways.
TNFR1 primarily mediates various biological effects through its interaction with its ligand, TNF-α. When TNF-α binds to TNFR1, it initiates a series of signal transduction events, including the activation of NF-κB (Nuclear Factor-κB) and the JNK (c-Jun N-terminal kinase) pathway. In the treatment of autoimmune and inflammatory diseases, researchers have developed biologics such as anti-TNF drugs (e.g., Infliximab, Adalimumab, etc.) to alleviate symptoms and slow disease progression. These drugs work by inhibiting the binding of TNF-α to TNFR1, thereby reducing inflammation. Additionally, TNFR1 is also implicated in tumor development and resistance, making it a potential target in the field of cancer therapy.
