HMGB1

HMGB1 (High Mobility Group Box 1) is a high-mobility group protein belonging to the chromatin structural protein family. It binds to DNA in a specific manner within the cell nucleus, assisting in the regulation of gene expression. The HMGB1 molecule typically consists of two DNA-binding domains (A box and B box) and a non-specific linker region (C box). HMGB1 serves multiple functions in cell biology. Within the cell nucleus, it binds to DNA, regulates gene transcription and chromatin structure, and participates in gene expression control. In the cytoplasm, HMGB1 plays a crucial role in cell stress, inflammation, and immune responses. When cells are damaged or infected, HMGB1 can be released into the extracellular environment, where it binds to specific receptors on immune cells (such as RAGE, TLR4), triggering immune cell activation and inflammatory responses. HMGB1 is regarded as an important proinflammatory cytokine that guides immune cell migration and activation, enhances inflammatory responses, and thus participates in immune responses and tissue repair. HMGB1 is also associated with the autophagy process and may play a role in autophagy regulation and execution. Research on HMGB1 has gained widespread attention in various fields, including gene regulation, inflammation biology, immunology, and oncology. IHC image of CSB-RA010553MA1HU diluted at 1:100 and staining in paraffin-embedded human salivary gland tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-Mouse IgG labeled by HRP and visualized using 0.05% DAB.

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
