Tau protein is a basic protein, as a member of the MAPs family of proteins, tau protein mainly acts on the distal end of axons to maintain the stability and flexibility of microtubules; tau protein interacts with tubulin to stabilize microtubules and drives tubulin Assemble within microtubules.. Tau protein controls microtubule stability through isomerization and phosphorylation.

Tau protein is very abundant in neurons of the central nervous system and relatively low in non-neuronal cells (astrocytes and oligodendrocytes, etc.). Tau protein is involved in regulating axonal transport and nuclear function to protect DNA integrity; it interacts with the actin cytoskeleton to promote actin filament formation, and regulates the NMDA receptor signaling pathway by interacting with Fyn. Phosphorylation of tau is regulated by a number of kinases, including PKN, serine/threonine kinases. The accumulation of hyperphosphorylated tau in neurons will cause neurofibrillary degeneration, which is associated with various neurodegenerative diseases such as AD and PD.
