Description

The ply polyclonal antibody production involves initiating an immune response in a rabbit using a recombinant Streptococcus pneumoniae serotype 2 ply protein fragment (encompassing amino acids 2-471) as the immunogen. Serum collected from the rabbit contains polyclonal antibodies. These antibodies are subsequently purified through affinity chromatography. Extensive validation procedures confirm the antibody's specificity for Streptococcus pneumoniae serotype 2 ply protein. This ply antibody is suitable for use in ELISA and WB assays.

The major virulence factor pneumolysin (ply), produced by Streptococcus pneumoniae serotype 2, functions as a pore-forming toxin. Ply forms pores within the cell membrane, which aid in the dissemination of bacteria and augment the disruption of cellular barriers and the demise of host cells, thereby playing a role in the development of the disease. It also exerts potent pro-inflammatory effects and participates in the invasion by breaching the epithelial layers.

Function
A cholesterol-dependent toxin that causes cytolysis by forming pores in cholesterol-containing host membranes. After binding to target membranes, the protein undergoes a major conformation change, leading to its insertion in the host membrane and formation of an oligomeric pore complex. Cholesterol is required for binding to host membranes, membrane insertion and pore formation; cholesterol binding is mediated by a Thr-Leu pair in the C-terminus. Can be reversibly inactivated by oxidation.
